Introduction: DNA, Deoxyribonucleic acid, is a double stranded, helical nucleic acid molecule which determines inherited structure of a protein. The"steps" are made of bases: adenine, guanine, cytosine, and thymine. The sides are sugar and phosphate molecules. Restriction enzymes are enzymes that cut DNA at restriction sites, leaving fragments blunt or sticky. The restriction fragments are separated using a technique called gel electrophoresis. DNA has a negative charge so when an electrical charge is applied it makes DNA move to the positive side. DNA is placed in agarose gel. Smaller fragments move faster. The purpose of this lab is to separate DNA fragments using gel electrophoresis. Hind III cuts AAGCTT between the two irst A's. EcoRI cuts at GAATTC between the G and the A. Hind III and EcoRI both make sticky ends. Results: Our results for this lab were EcoRI separated into five fragments. Hind III separated into four fragments. The control only had one fragment. (See chart A and figure 1-1 for distances) Discussion: The purpose of this lab was to see how gel electrophoresis separates DNA fragments. We used Hind III, EcoRI, and a controlled enzyme. Some fragments were hard to see because of smearing. These were the bigger fragments. Loading the DNA was difficult and if you weren't careful you could rupture the wells which ruined the lab. We, fortunately, did not run into this problem. Abstract: The purpose of this lab is to separate DNA fragments with gel electrophoresis using EcoRI and Hind III. Restriction enzymes are used to break up the DNA, then negatively charged DNA is placed in a gel casting tray. Then it is placed into an electrophoresis chamber. An electrical field is placed across the agarose gel which forces the fragments to move down the gel. The amount of lines show how many fragments there is in the DNA. We had five fragments for EcoRI and six for Hind III. The no enzyme had only one fragment. Procedures: We sealed the ends of a gel casting tray with masking tape and inserted a comb into the slots. The tray was filled about 6mm high with agarose gel. It covered half the height of the comb. We waited ten minutes for the gel to solidify. Then we placed the tray in a gel box and made sure that the comb was at a negative (black) end. The box was filled with tris-borate-EDTA buffer so it covered the entire surface of the gel. The combs were removed without ripping the wells. The micro pipet was used to load the lambda EcoRI, lambda Hind III, and lambda only into the wells. We dipped the pipet trough the surface of the buffer over the wells and expelled the contents. The top of the electrophoresis chamber was closed and electrical leads were connected. The dye was observed as it moved shortly after the power supply was turned on. The power supply was turned off after the bands migrated near the end of the gel and the top of the electrophoresis chamber was removed. We removed the gel from the gel casting tray and examined it under a light box and compared it to the ideal gel (figure1-2). Knurling  is a process of impressing a diamond-shaped or straight-line pattern into the surface of a work piece by using specially shaped hardened metal wheels. Ex. â€Å"Walnut Knurled Guitar Knob.† â€Å"Solid  walnut knurled  legs on table and chairs.† Two foreign borrowings, Knesset and knish, do not belong to the category of words spelled with the phonogram kn. They are spelled with the phonogram k; the sound /k/ is pronounced at the beginning of these words. Knesset (noun): The parliament of the State of Israel. The word derives from a Hebrew word meaning â€Å"a gathering.† Ex. â€Å"On July 11, 1995 this problem was raised for discussion in the Knesset finance committee.† knish (noun): A dumpling of flaky dough filled with chopped liver, potato, or cheese, and baked or fried. However, still having the device, Gardener was able to rape and murder two helpless teenagers back in 2009. It is evident that the paroled felon was not at any chance under careful supervision as officials thought. It can be depicted that these two victims could be alive if only the devices were effectively used. As a dangerous sex offender, John Albert Gardener still roams in the streets.

The Lack of Man in Lac Léman Essay -- Literary Analysis

The narrator of Hubert Aquin’s novel Next Episode cannot breathe; he’s drowning. The author presents water as a deadly force that has the ability to stifle life. Aquin employs the characteristics of water to represent the narrator’s inability to express his national identity and therefore himself. Multiple symbols are used throughout the book, such as fire and ice, but water is an overpowering and important aspect of the book because it conveys a sense of inertia in the individual. This notion of water is connected to the repression of Quebec nationality. The book discusses water in a variety of different contexts: as the primary foundation for the story (as interpreted by the translator), as a metaphor for self and as a replacement for language. Water, weaved into a nationalistic allegory, is given conflicting attributes and is both a life giver and life taker simultaneously. This double approach to the basic element of water embodies both his drive to express h im nationality and his incapacity to authentically vocalize his own existence. The tale begins below the waters of Lac Là ©man, as the narrator states â€Å"it’s in the area of this invisible lake that I’ll set my story† (4). The lake is central to the story, as it is this water (or water which flows from it) that he speaks of in terms of the self and language. The place names of this body of water are also significant for French-Canadian nationality. The translator Sheila Fischman has as much literary power as the narrator. This is because without having read the French edition of the text, the English edition modifies meaning concerning the image of water. Fischman does this by consistently using the French name for the lake â€Å"Lac Là ©man† instead of using its English name â€Å"Lake ... ...erves to corroborate the asphyxiation of a definitive French-Canadian national narrative. Water at once embodies the narrator’s want to express his nationality and personal identity, but also his inability to do so. Ultimately, Water becomes a type of liquid courage for the narrator that never produces substantial results, so he looks towards a future revolution to conceive a genuine national identity. Media Reaction Paper: West Side Story West Side Story was one of the first musicals this writer saw on Broadway. It was such a mesmerizing experience to live the culmination of the hard work performed by a group of very talented individuals that stemmed from a single idea. It was not until research was done for this paper that this student realized what a great cultural impact West Side Story was when it was first introduced to audiences. The book for West Side Story was written by Arthur Laurents in 1949. An interesting fact about this legendary story is that it was not always named West Side Story. The original name given this tale, based on Romeo and Juliet, was â€Å"East Side Story. † It was written as a rivalry tale between a gang of Roman Catholic teenagers named The Jets, and a gang of Jewish teenagers named The Emeralds. Because this topic of rivalry between the Jewish and Roman Catholic communities was redundant in the industry at the time, the creative team composed of Arthur Laurents, Jerome Robbins and Leonard Bernstein, decided to put the project on hold. Thus, they ceased work on the idea for a period of almost five years (Miller, 2006). After five years the creative team reunited to work on other projects and decided to revisit East Side Story and give it a completely new twist and decided to set it in Harlem, New York City (Miller, 2006). West Side Story depicts the love between a Caucasian teenage boy from the West Side of New York City and a young Puerto Rican girl from the same neighborhood. The characters were all from a poor neighborhood and hung out on the streets together. There were two gangs that were in constant conflict with each other: The Jets and The Sharks. Tony, the main character, is a member of the Jets, a gang composed of Caucasian teenage boys. He is however, different from the rest of the members and wants to work and someday go on his own way. Maria is the sister of the leader of the Sharks, a gang composed of teenage Puerto Rican boys. Maria and Tony meet and instantly fall in love and have to deal with the turmoil of being from different cultural backgrounds and the difficulty of having their love accepted by each of their groups. The relationship causes conflict in the neighborhood that results in violence and ultimately Tony’s death. West Side Story was considered a breakthrough for the Puerto Rican community. Very little exposure about the Puerto Rican culture and their presence in New York City had been portrayed in any type of media up to the time the play’s debut on Broadway. Although Puerto Rican’s were depicted with stereotypes that were not necessarily true about their culture, the story delivers a message of change. It describes racism as dangerous and as an obstacle to love. To have developed such a controversial story during times when segregation still existed in the United States was very brave. What softened the diversified and strong topic was the fact that it was a musical with very vivid choreography and colors andan upbeat script full of humor. This factor made the story more acceptable to the American people. The media has its ways to persuade the masses. Within the past century, the media has played an important part in the cultural changes in America. Television, radio, theater and written works have broken through the racism barriers to expose the many faces of the American population. Everyone is different and their cultural background makes a special contribution to the community of the world. West Side Story is a good example of a media piece that addressed America’s cultural diversity in a time when most people had no idea how multicultural our country really was. References Miller, G.
